چکیده

Lipid peroxides in tissues were fluorometrically determined using 1,3-diphenyl-2-thiobarbituric acid (DPTBA) as a fluorogenic reagent, and the results were compared with those obtained by the DPTBA-visible method, thiobarbituric acid (TBA)-fluorescence method, or TBA-visible method. The optimum conditions for determining lipid peroxides were analyzed by using brain homogenate. The optimum pH for the reaction of brain homogenate with DPTBA was 2.5. The amount of DPTBA suitable for the reaction with 100 μl of a 1 % brain homogenate was 0.12 M. A linear relationship was observed between the fluorescence intensity and the amount of malondialdehyde (MDA) in the range of 1.25-12.5 nmol/ml. The detection limit of the DPTBA-fluorescence method was 0.006 nmol/g wet weight at a signal-to-noise ratio of 3. The sensitivity of this method was 5.5 times higher than that of the TBA-fluorescence method, lipid peroxides in tissues could be easily determined without separation from

Fluorometric analysis of 2-thiobarbituric acid reactive substances in turkey.

Three experiments were conducted to develop a sensitive and reliable fluorometric thiobarbituric acid (TBA) reactive substances (TBARS) method to determine lipid oxidation products in meat. The first study was conducted to find the optimum sample preparation conditions for meat in the fluorometric method. The second study was to compare the original and the modified methods by using meat and bl...

Inhibitory effect of citric acid on rancidity of frozen catfish (Silurus glanis)fillets

The effect of aqueous solution of citric acid (CA) on the lipid stability of the European catfish (Silurus glanis) fillets during frozen storage (up to 6 months) was investigated. Rancidity development was measured by several biochemical indices (free fatty acids, peroxides and TBA) and complemented by the measurement of expressible moisture and sensory evaluation (flesh odor, consistency and f...

Protective Effects of Tormentic Acid, a Major Component of Suspension Cultures of Eriobotrya japonica Cells, on Acetaminophen-Induced Hepatotoxicity in Mice.

An acetaminophen (APAP) overdose can cause hepatotoxicity and lead to fatal liver damage. The hepatoprotective effects of tormentic acid (TA) on acetaminophen (APAP)-induced liver damage were investigated in mice. TA was intraperitoneally (i.p.) administered for six days prior to APAP administration. Pretreatment with TA prevented the elevation of serum aspartate aminotransferase (AST), alanine...

Solid-phase microextraction method for the determination of volatile compounds associated to oxidation of fish muscle.

A procedure for the determination of volatile compounds derived from lipid oxidation of fish muscle samples is presented. Analytes are concentrated on a solid-phase microextraction fiber employed in the headspace mode (HS-SPME), and selectively determined using gas chromatography in combination with mass spectrometry (GC-MS). The influence of several parameters on the efficiency of microextract...
